RFD4-Proteina guide demonstrates protein design in a browser
The guide demonstrates protein design using RFD4-Proteina in Hugging Face Spaces, with the target protein PD-L1 as an example. NVIDIA covers access to dedicated GPUs from 21 October to 4 November 2026; the shared ZeroGPU environment also remains free, subject to quotas.
The Hugging Face blog published a guide to protein design using RFD4-Proteina, a model jointly developed by NVIDIA and the Institute for Protein Design at the University of Washington. The model jointly generates protein sequences and all-atom structures. It can be tried in a browser without installation through two Hugging Face Spaces environments featuring the AtomWorks Studio molecular viewer and editor.
The guide uses a prepared example with the target protein PD-L1. The input includes the target structure, the desired binding region, and specific atoms or amino acid residues marked as desired contact sites, known as hotspots. After generating a design, the user checks contact with the marked sites, records the seed, and compares additional variants while keeping the input unchanged. Designs, the annotated target, and complexes can be downloaded in CIF format.
The thinking setting offers low, medium as the default, and high levels. According to the article, a higher level increases the likelihood of meeting computational criteria, but also increases computing resource consumption. A generated design does not by itself confirm actual binding to the target; this must be verified experimentally.
The dedicated environment runs on an NVIDIA RTX PRO 6000 GPU, and NVIDIA covers access from 21 October to 4 November 2026. After that, this environment will no longer be free. The alternative ZeroGPU environment uses shared capacity and remains free within the account quotas on Hugging Face.
Why it matters
Researchers can start with a specific binding target and obtain both a sequence design and a three-dimensional structure. Exporting allows them to follow up with their own computational checks, such as assessing thermal stability or the tendency to aggregate, and select candidates for experimental validation.
Two audiences, two different impacts
What this means
For individuals
An individual can use the prepared PD-L1 example to try specifying a binding target and comparing designs without installing the model.
For a business
For a research team, the conditions for accessing computing capacity change after 4 November 2026: funded access to the dedicated environment ends, while the free shared option is subject to account quotas.
